Entrada Therapeutics, Inc. is a clinical-stage biopharmaceutical company in Boston developing intracellular therapeutics on its Endosomal Escape Vehicle (EEV) platform. An EEV is a cyclic peptide that binds the cell's phospholipid bilayer, triggers endocytosis, then escapes the endosome so a conjugated oligonucleotide or protein reaches the cytosol. Its lead franchise is four EEV-PMO exon-skipping conjugates for Duchenne muscular dystrophy, led by ENTR-601-44. That candidate's IND was on clinical hold until Entrada resolved it in February 2025. In the second quarter 2026 report, Cohort 2 of the Phase 1/2 ELEVATE-44-201 trial was fully enrolled with topline data due in the first quarter of 2027, and ELEVATE-45-201 Cohort 1 data due in October 2026. Under a collaboration and license agreement with Vertex Pharmaceuticals, Vertex holds an exclusive worldwide license to VX-670 for myotonic dystrophy type 1; Entrada received $223.7 million upfront plus a $26.3 million equity investment and is eligible for up to $485.0 million in milestones. Collaboration revenue was $25.4 million in 2025 against a $143.8 million net loss, and $1.8 million in the first half of 2026 against an $82.5 million loss. Cash and marketable securities were $223.0 million at 30 June 2026, expected to fund operations into the third quarter of 2027. Entrada had 152 full-time employees as of 19 February 2026.
